Am I better off glitching Hell Countess?

I'm primarily interested in runes and keys. Am I better off glitching Hell Countess than completing the quest?

Nope. You don't gain anything from glitching the countess because she only has a normal drop, and not a special quest drop like the act bosses.

Doesn't the chest only open when you complete the quest? How does the drop from the chest compare with the drop from the Countess?

Aye, the chest only opens if you complete the quest. The chest in the countess area is pretty much just a normal "special chest", with the addition of a bunch of gold dropping as well. In other words, if you wanted a drop from a special chest then Kurast or Pit might be quicker.
