I read that if you kill a Boss and close the quest for him, the next time you kill him his drops will not be very good.
Is it a very significant difference in drops?
As in, is it not worth it at all to repeat a Boss if you finished the quest to kill him?

It's definitely still worth it run bosses.

Meph drops a lot of good mid-level gear (skin of the vipermagi, goreriders, war travs, harley, etc...). He's your first stop if you're just starting out.

Andariel is the exception due to a bug, she always drop like it's your first kill. She's particularly good for jewelry.

Baal can drop any item in the game with the obvious exception of the anni and hellfire charms.

First-kill chances of unique, set and rare are higher, plus the no-drop chance is nonexistant. But that doesn't mean the subsequent kills are useless. Most people find their items from doing lots and lots of Meph, Baal or Chaos runs.

Yeah but I was trying to figure out if those people where 'bugin' the drop rate entering games where they cant finish the quest or not...Thanks.

Some people bug the quest so that they always get the quest-drop but it's not necessary at all. Besides if you 'bug', for example, Mephisto, you won't be able to join any dia- or baalruns, which means you'll MF on lower level and thus you may miss some synergy bonuses for your skills etc.

no-drop chance still exist in first drop kill, actually it's even little bit higher (because junk drop is excluded from dropping, all other options have higher chance to occur - equippable items as well as no-drop).

Chance to get a unique item is around 1.5 * better on first kill compared to regular kills.

Well. you can run mephisto as firstkill for a while and when you are bored move on to Pindle, Diablo and Baal.

You will also have tunnels for blizzard sorcs and pits for other characters.
