mf affects on the items dropped that are normally white to be either blue, yellow, or even gold.
the number of players in the game affects countess though- more players in the game will mean less keys or runes that could drop, I believe

The countess has rune drops and non-rune drops. The non-rune drops can be runes though. Infact the highest rune she can drop is only as a non-rune drop. The more players the more likely the non-rune drop will be something other than a rune.

More people in the game does increase the odds of a monster dropping an item, wether it be a rune or any other item. If you can kill the countess with more people in the game just as easily then I say do it. If it slows you down then you might just want to take all your runes and upgrade them to the one you want using the cube. Cubing may sound like a pain, but it is safer than dying and probably quicker than trying to kill her with more people in the game and still getting low runes. Good luck on whatever runeword you are trying to make.
As for hellforge I'm not sure I haven't done any rushes or had a lot of people in the game when I did that quest. I would believe that the hellforge would act like a chest and drop more runes/gems with more players in the game. Not sure if they have to be in the same area or getting the quest at the same time.
As for the part about non-rune drops. I've never heard of this I thought that a rune was considered an item and that she just dropped them most often because of her good drop chances. I guess that blizzard might have made it so that she has more drops checks. A few for runes and another for non-runes.

There are writeups about countess rune drops. I"m too lazy to find the links. Basically it explains better than I do about the rune drops and non-rune drops and why she drops fewer runes on average with more people in the game.
and at least one tells you how many times on average you will have to kill the countess to cube a zod.

MF never has an effect on rune drops.
Number of players does not affect forge drops --- however, do keep in mind that there is only ONE rune per game, not one rune per player getting the quest. So even if 8 people are getting the quest, only one rune will drop.
The countess has a special rune drop, which can only happen for "no-drops". As more players in the game reduce the chance for a no-drop, more players will mean fewer special rune drops.
The special rune drops are capped well below what the countess can drop with her normal drops, though --- so having more players means more normal drops, which means higher chance of getting one of her "better" runes, just like with all other monsters.

As far as I know, the terror key is part of the countess's item drop, not rune drop. So, the more players in the game, the better.
The countess has a special rune drop, which can only happen for "no-drops". As more players in the game reduce the chance for a no-drop, more players will mean fewer special rune drops.
Just to add, the countess can only drop a maximum of 6 items. Her item drop can drop up to 5 items, and the rune drop can drop up to 3 items. After she is killed, the game first performs her item drop, and then her rune drop. Since a higher player count game decreases the chance of a "no-drop", then there's a greater chance of her dropping all 5 items from her "item drop" set, which means she will only be able to drop a single rune from her "rune drop" set.
Ideally, if it's runes your after, you'll want to make sure she always has a chance to drop all 3 runes from her rune drop. This means having a lower player count game, as to increase the chance of a no drop from her item drop set.
