Hiyas. Firstly, welcome to the Forums. Hope you'll have a great time here :)
Secondly, with regards to hotkeys, I assume you want to assign skills to them. If so, move your mouse pointer over the skill box - it's the box which is located on the bottom left and right of the screen. I think the default is attack which is the one with an arrow pointing downwards.
Once you do so, more boxes will pop up. You can then move your mouse pointer over the selected box which you want to assign a hotkey to. Each box represents a skill which your character has. It even includes casting town portal or identify (if you have the tomes/scrolls in your inventory) or certain other skills/spells which may sit in items (eg. a staff with 20 charges of nova).
So, once you've moved your mouse pointer over a skill box which you want, press the hotkey you want to assign to it, eg. F4, F8, G etc. The next time you need to change skills to that, pressing the selected hotkey will immediately select that skill.
On the other hand, if you wanted to assign hotkeys to generic skills like running or standing still, it's in the options section. Press ESC, you'll see Game Options (I think) and then you'll get to select Configure Controls (I can't remember the exact words). You'll see a list of generic skills and the default hotkeys which are assigned, eg. Map = TAB, standing still = SHIFT. You can change these around.
However, what I've realised is that these changes are character-specific (well, on battle.net anyway). So, if you're playing Character 1 and you've changed his hotkeys, when you move to Character 2, your changes won't apply. You'll need to make the same changes again.
Hope that helps. If you still need help, let me know :)

They are character-specific on SP as well.
